Gertrude Trudy ; Elion's career has important lessons for all who strive to join the constant battle against disease. Intellectual modesty, perseverance, dedication, sensitivity to her co-workers, courage, integrity, and above all, focus were the hallmarks of her style. Trudy was preeminently goal-oriented and her goal was drug discovery. This is not to say she did not possess wide scientific interests, but she had remarkable discipline and focus in using her laboratory time and resources. If a line of inquiry, however interesting, was not on the path to drug discovery, she simply said no. Trudy would never argue that there wasn't a legitimate place for "science for science's sake"--it just was not for her. Her compassionate nature and personal experiences made very real to her the vast number of people hoping for a remedy to their diseases. Many scientists lose sight of the potential human impact of their work in the insular environment of the laboratory. She did not, and had a sense of urgency about it, never letting anything else get in the way. Drug discovery was her mission. Trudy didn't agonize over her work--she enjoyed it immensely. Her favorite activity was to analyze data and then to fit the resulting piece of new information into the complex labyrinth that leads to a new drug. She had the patience and discipline to proceed one step at a time, but each step was carefully calculated. She derived as much satisfaction from the quest as from the final achievement. Trudy's courage was impressive. She was a "can do" person, but was never overly optimistic. She entered new fields with gusto when the quest led her to them. George Hitchings, her mentor first and then her long-time co-worker, always used the term "quick learn" to describe Trudy's ability to go into a new field, quickly master it, and then make important contributions. Another aspect of her courage was her readiness to speak out for what she felt was just and right. On occasion, she argued with the FDA, with the management of Burroughs Wellcome Co., and with the Wellcome Trust. Yet she did not seem to make enemies, perhaps because the targets of her outspokenness ultimately respected her integrity. Trudy's enthusiasm was contagious. She was a team builder gifted with the ability to be critical and supportive simultaneously. She helped individuals find where they could best contribute and was always generous with credit. Her personal warmth and sensitivity were pervasive in her dealings with her co-workers. Trudy was always clear about what she knew well and what she did not, so establishing outside collaborations was standard procedure for her. In these collaborations, she quickly established trust and mutual respect--often making lifelong friends. After receiving the Nobel Prize, she was asked to co-chair a World AIDS Conference with Mother Teresa. She declined because she felt that Mother Teresa deserved to be the sole chairperson, yet Trudy probably did as much as anyone to alleviate human suffering. The sales volume of each of the antimicrobials supplied in the year 2001 9 ; was defined as the antimicrobial usage in each species of animal cattle, pigs, broiler chickens, and layer chickens ; . As regards the -lactam antibiotics, the volume of usage of the penicillins i.e., the sum of aspoxicilin, amoxicillin, ampicillin AMP ; , cloxacillin, benzylpenicillin, and mecillinum ; and cephalosporins the sum of cefazolin, ceftiofur and cefquinome ; were calculated separately. The sum of the volumes of tetracycline, oxytetracycline OTC ; , doxycycline, and chlortetracycline usage was deemed as tetracycline usage, since OTC-resistant bacteria exhibit cross-resistance among these antimicrobials 10 ; . As regards the aminoglycoside antibiotics, the volumes of use of kanamycin KM ; , gentamicin GM ; , and streptomycin including dihydrostreptomycin [DSM] ; were divided, as isolates resistant to one of these agents are not consistently cross-resistant to the other agents 10 ; . Several factors must be examined to make such determinations including, but not limited to, the volume of antibiotics used, the fate of antibiotics used, the relative impact and ecological significance of antibiotics on microbial flora in specific environments, and the probability that use of antibiotics under aquaculture processes leads to development of human pathogens resistant to that antibiotic. Scientifically credible data has been scarce leading to considerable speculation. This report provides information as to the actual volume of antibiotics active ingredient A.I. ; sold to the domestic aquaculture feed industry in 2001 and 2002. It is assumed that the volume sold is equivalent to the volume used that year in the domestic aquaculture industry. Only two antibiotics are approved for use by the FDA Center for Veterinary Medicine CVM ; and available in the domestic aquaculture industry. These are approved only for therapeutic purposes for catfish, salmonids, and lobster oxytetracycline ; or for catfish and salmonids ormetoprim: sulfadimethoxine ; . In the 25 month period from Jan. 2001 to Feb. 2003, there was 36, 126 kg of ormetoprim: sulfadimethoxine A.I. sold. Averaged out, the amount sold per year was 17, 340.48 kg of ormetoprim: sulfadimethoxine A.I. In 2001 there was 15, 200 kg A.I. of oxytetracycline sold and in 2002, only 7, 134 kg A.I. sold to the domestic aquaculture industry. The combined total is 32, 540.48 kg A.I. 71, 752 lbs ; sold in 2001 and 24, 475 kg A.I. 53, 967 lbs ; sold in 2002. It has previously been speculated that co-tolerance could be more prominent among bacteria than among higher species, due to the ease of transfer of genetic elements and to the colocation of resistance genes on mobile genetic elements such as plasmids Blanck et al. 1988 ; . The level of resistance to macrolides in strains isolated from farm animals or slurry samples is sometimes in the same range as tetracycline resistance Cotta et al. 2003 ; , and sometimes lower Hayes et al. 2003 ; . Further, macrolide and tetracycline resistance genes sometimes co-reside on conjugative transposons found in Bacteroides species Courvalin & Carlier 1986, Caillaud et al. 1987 ; . Compared with bacterial isolates from farm animals, the presence of resistant bacterial strains in environmental compartments not impacted by antibiotics is controversial: whereas some investigators report a lower frequency of resistance in remote areas Esiobu et al. 2002 ; , some studies find highly resistant bacteria also in situations without antibiotic pressure Stanton & Humphrey 2003 ; . Thus, it is difficult to say whether the limited co-tolerance observed in our study is due to a limited occurrence of easily transferable genetic elements with multiple antibiotic resistances or due to other reasons, such as a low transfer frequency or a predominance of other tolerance mechanisms. In our study, co-tolerance between different compound classes has been observed in one soil microcosm: the highest concentration of tylosin caused an increase in the oxttetracycline tolerance as well. As this effect only occurs with the highest concentration of tylosin, this cotolerance might be caused by selective depletion of the community at that concentration and the intrinsic tolerance of the remaining soil bacteria. Tylosin is indeed known to have selective activity: it belongs to the narrow-spectrum macrolides that affect mostly Grampositive cocci and bacilli as well as Gram-negative cocci Nakajima 1999 ; . Oxytetracycline, in contrast, is a broad-spectrum agent Chopra 2001 ; . Our results confirm other studies that showed a limited co-tolerance for structurally unrelated compounds, for example for different uncouplers and photosynthetic inhibitors Blanck & Molander 1991, Blanck & Wngberg 1991 ; . On the other hand, co-tolerance has been frequently observed for metals, for example between copper, cadmium and zinc, as observed in field and microcosm experiments Daz-Ravia et al. 1994, Gustavson & Wngberg 1995, Bth et al. 1998, Soldo & Behra 2000 ; . Although metal co-tolerance has often been described, the reasons for this phenomenon are not generally agreed upon. The same holds true for the co-tolerance observed between two structurally unrelated compounds, diuron and TBT, in periphyton communities Molander et al. 1992 ; . Overall, the small co-tolerance between different antibiotic agents was small. However, this does not relieve the notion that co-tolerance might be a limiting factor for the application of PICT to investigate the relevance of a certain toxicant in a given system.
