Phosphorylation of the purified sArr will be characterized in reconstitution assays with rhodopsin and rhodopsin kinase. pDEST26-sArr was transfected into human embryonic kidney HEK293T ; cells along with muscarinic acetylcholine M1 receptors and invertebrate Gq. The function of arrestin was determined by Inositol 1, 4, 5-triphosphate IP3 ; formation assay. In HEK293T cells, carbachol-stimulated IP3 formation was reduced in sArr-transfected cells over 1-hour stimulation. According to modern Western medicine, irritable bowel syndrome IBS, also known as spastic colon, functional or nervous diarrhoea, colon or vegetative neurosis, or mucus colitis ; , is an non-inflammatory bowel disease. Aetiology The causes of IBS are not entirely clear. Apart from motor-secretory activity of the colon due to excessive parasympathetic stimulation, there is no evidence that any structural defects of the bowel are associated with IBS. However a variety of factors have been identified as possible causes: psychological such as family, marital or work problems, tension and stress. irregular diet such as a low-fibre diet. food allergies such as intolerance to wheat, corn, dairy products, citrus fruit, tea, coffee, apples, pears and salads. Incidence IBS is the most common gastrointestinal disorder reported to GPs in Western industrialised societies; 30-50 % of all patients referred to gastroenterologists suffer from the condition. However it is almost impossible to determine the exact number of sufferers as many do not seek medical attention. Some estimates suggest that approximately 15% of the population have suffered from IBS, and that there are twice as many women sufferers in the 20-40 age group ; as men, although it is more likely that the number is equal, with men not reporting the symptoms as often. Causes: There are no mechanical, biochemical or overt inflammatory causes of IBS. However, altered gastrointestinal GI ; motility and increased gut sensitivity are thought to play a causal role. Some studies reported increased bowel contractions temporally associated with abdominal pain; heightened sensitivity to visceral distension has also been described. These are similar to the GI effects of stress reported from human and animal studies. Symptoms include pain, diarrhoea associated with urgency, anxiety and rarely faecal incontinence ; or constipation associated with bloating discomfort and an altered body image ; . Diagnosis is based on the presence of validated symptoms, absence of atypical IBS symptoms and a normal physical examination. Limited relevant diagnostic testing may be used to reassure patients. Non-pharmacologic therapies alone are often adequate for many patients. Identification of potential triggers is important for lifestyle modification. Special diets are not sufficient to eliminate symptoms but avoidance of identifiable dietary excesses, caffeine and any dietary triggers will help reduce attacks.
